I don't see the TangiBot as damaging to makerbot or the RepRap cause.
I think that anything delivers 3D printing into the hands of more people pushes the community forward.
A clone of an existing product is not the most "glorious" way to make it happen; but hey, it will probably work.

It would be very nice to see a 3D scanner and/or filament extruder become popular and supported by the community
I think that the RepRap will not be able to achieve the status of philanthropic wonder machine until a plastic recycler (filament extruder) is viable.

It is a bit vanilla, but Spacxula's calibration set is where I started

I like to use the 20mm block; I just print the first layer to make sure my "width over thickness" is on the money.
Maybe others would like to post their favorite calibration items here

The 200% on the first layer is new (as far as I can tell)
I also think that, setting the value to 0 makes it default to 200% also.
I manually enter my values usually, but gave it a shot as it was just for kicks; it did not work so great.
I then manually entered values for the widths and was up and printing great.

It is great in fact that there are multiple efforts out there right now.
My first successful prints came from skeinforge too.

Hmmm,
On a ten foot tall machine, I am thinking some kind of chain or monster rack and pinion.
But the rack and pinion would limit the speed of the machine for sure.

Well,
Be sure to calibrate the steps per mm for your extruder; ie. make sure a command to extrude 100mm moves 100mm of filament exactly.
Then experiment with your width over thickness so that the lines of extruded plastic are touching.
I like to print the first layer of a 20mm cube; this let's me make sure I am printing a perfect little "postage stamp"
I want that stamp to be perfectly smooth

But ditch skeinforge and get onboard the Slic3r bandwagon.
Once again not an impartial observation...
But Slic3r is better, faster, stronger. It has precompiled versions, slices models in seconds, new releases monthly, an easy to use GUI... The list goes on.
Skeinforge is better at making support structure. PERIOD.

It is an exciting time for sure
I am wondering if Delta Bots will take over the fused filament scene; I actually kind of hope so.
I feel like the giant heavy Cartesian machines are more in line with traditional CNC as opposed to FFDM which should need no force to move about.

I am intrigued by your argument that the end effector has to be made by the machine itself; However, I think there will be plenty of folks out in TV land with non-printed hotends/extruders who probably think they own a RepRaps
